چکیده
Square-wave cathodic adsorptive stripping voltammetry based on adsorptive accumulation is a very sensitive technique for the trace determination of xanthosine 50-monophosphate (50-XMP) and xanthosine 50-diphosphate (50-XDP). The determination is based on the strong interaction of the adsorption of xanthosine phosphate compounds on a mercury electrode surface, forming Hg(II)-xanthate. The cathodic reduction of the accumulated Hg(II)-xanthate complex provides the basis for direct stripping measurements of the investigated biological compounds at subnanomolar concentration levels. Moreover, controlled adsorptive accumulation of the Cu(II) complex of xanthosine phosphate is also reported to assay trace amounts of xanthosine phosphate. The height of the sharp chelate peak of adsorbed Cu(II)-xanthate, coupled with the flat baseline, facilitates measurements at nanomolar and submicromolar concentration levels. The adsorption and the redox behaviour of the investigated complexes are indicated by cyclic voltammetry. Experimental and instrumental parameters for the quantitative determination were optimized. Statistical analysis of the calibration curve data is also included.

Differential pulse and square-wave cathodic stripping voltammetry of xanthine and xanthosine at a mercury electrode.
The surface activity of xanthine (Xan) and xanthosine (Xano) at a hanging mercury drop electrode (HMDE) was studied using out-of-phase ac and cyclic dc voltammetry. The results show that Xan and Xano were strongly adsorbed and chemically interacted with the charged mercury surface, which is the prerequisite step for applying the cathodic adsorptive stripping voltammetric determination of such b...
متن کاملStructural and functional characterization of a noncanonical nucleoside triphosphate pyrophosphatase from Thermotoga maritima.
The hyperthermophilic bacterium Thermotoga maritima has a noncanonical nucleoside triphosphatase that catalyzes the conversion of inosine triphosphate (ITP), deoxyinosine triphosphate (dITP) and xanthosine triphosphate (XTP) into inosine monophosphate (IMP), deoxyinosine monophosphate (IMP) and xanthosine monophosphate (XMP), respectively. The k(cat)/K(m) values determined at 323 and 353 K fall...

The levels of endogenous caffeine and theobromine were much higher in buds and young leaves of Coffea arabica L. cv Kent than in fully developed leaves. Biosynthesis of caffeine from 14C-labeled adenine, guanine, xanthosine, and theobromine was observed, whereas other studies (H. Ashihara, A.M. Monteiro, T. Moritz, F.M. Gillies, A. Crozier [1996] Planta 198: 334-339) have indicated that there i...
متن کاملFormation of an adenyl xanthosine monophosphate intermediate by xanthosine 5'-phosphate aminase and its inhibition by psicofuranine.
The amination of xanthosine 5’-phosphate and its inhibition by psicofuranine was examined with substrate amounts of purified xanthosine St-phosphate aminase from Escherichia coli. In the absence of NH,, incubation of the enzyme with adenosine triphosphate-8-i4C and XMP-8J4C for 10 min results in the conversion of ATP to AMP without concomitant formation of guanosine monophosphate. Shorter perio...
متن کاملPlant Purine Nucleoside Catabolism Employs a Guanosine Deaminase Required for the Generation of Xanthosine in ArabidopsisW
Purine nucleotide catabolism is common to most organisms and involves a guanine deaminase to convert guanine to xanthine in animals, invertebrates, and microorganisms. Using metabolomic analysis of mutants, we demonstrate that Arabidopsis thaliana uses an alternative catabolic route employing a highly specific guanosine deaminase (GSDA) not reported from any organism so far. The enzyme is ubiqu...
